FLLL32是姜黄素的小分子类似物,它可以抑制STAT3的信号转导和活化。在pSTAT3阳性的A375细胞系,FLLL32导致促凋亡作用,48小时的IC50值为1.3 μM[1]。
在黑色素瘤中,STAT3的激活与预后不良相关。STAT3能促进血管生成和细胞增殖、推动侵袭和转移,抑制凋亡[1]。
在多种人类黑素瘤细胞系中, FLLL32处理24小时之后,抑制了STAT3 Tyr705位点的磷酸化,但对于Tyr727没有抑制作用。在体外细胞实验中,FLLL32可抑制Jak2的活性。但在多种黑色素瘤细胞系的实验中,即使在8 μM 浓度的FLLL32下,Jak2的磷酸化没有明显的改变。这意味着FLLL32可能直接作用于Stat3蛋白。2-4 μM 的FLLL32作用仅4小时就减少了pSTAT3并诱导细胞死亡[1]。
在皮下异种移植B16F10黑色素瘤的C57BL/6小鼠模型中,与DMSO处理的动物相比,每天腹膜内给药25 mg/kg和50 mg/kg的FLLL-32显著的延迟了肿瘤生长(P <0.001)[2]。
