FIIN-2是FGFR1-4的不可逆抑制剂,IC50值分别为3.1\4.3\27和45 nM.FIN-2也能适度抑制EGFR,IC50值为204 nM.FGFR2的Cys491是FIN-2结合的主要标记位点[1].
FIIN-2是一类抑制剂,依赖于FGFR1或FGFR2的看门突变而高效抑制细胞增殖.已有报道FGFR4和FIIN-2的共结晶结构,其意外地具有”DFG-out”共价结合模式.FIIN-2保留FIIN-1的嘧啶[4,5-d]嘧啶酮核心,但是移去了对FIIN-1高效抑制FGFR非常重要的两个氯原子.FIIN-2维持共价结合P-环半胱氨酸的能力,但是改变了对其它激酶的选择性.与FIIN-1相比,FIIN-2有力结合FGFRs,表现出良好的总体激酶选择性.FIIN-2对EGFR亲和力较低,但是FIIN-3高效结合野生型EGFR及其一系列突变体[1].
FIIN-2抑制FGFR1-4 Ba/F3细胞增殖,EC50值在纳摩尔范围内,其特别高效地抑制FGFR2,EC50值在1 nM范围.FIIN-2也能抑制FGFR2 V564M看门突变的Ba/F3细胞,EC50值为58 nM,而FIIN-1和BGJ398对此突变的EC50值高于1.0 μM.FIIN-2高效抑制看门突变体V564F.相比之下,FIIN-2的效力要减少四倍(EC50为506 nM).FIIN-2在浓度高达1.8 μM时对EGFR L858R无活性.FIIN-2对蛋白激酶FLT1(FLT1)的抑制作用比较弱.在野生型FGFR2 Ba/F3细胞中,FIIN-2完全抑制FGFR2突变体酪氨酸656/657和V564M的自磷酸化,分别在浓度为3 nM和300 nM时[1].
