人表皮生长因子受体(ErbB2)是一种跨膜受体,在15%到25%的乳腺癌细胞中过度表达。ErbB2的过度活跃造成不利的生物学特性和不良的临床结果1. ErbB2在细胞系中的过表达导致在没有配体情况下的转化2。EGF家族的配体不直接结合ErbB2。因此,从技术层面上说,ErbB2仍然是一个孤儿受体。
多种人类肿瘤中含有扩增的或过表达的ErbB-2基因,编码生长因子受体蛋白。在NIH / 3T3细胞中,当ErbB2的互补DNA 在SV40启动子的控制下表达,可检测到ErbB2蛋白质的表达,但基因仍缺乏转化活性。莫洛尼鼠白血病病毒的长末端重复序列可以使ErbB2蛋白的表达增加五到十倍,与ErbB2蛋白的激活相关,是一种强效致癌基因。高表达的ErbB2产物与NIH/3T3细胞恶性转化相关,在过表达该基因的人类乳腺肿瘤细胞中也有这种现象。
