表皮生长因子受体(EGFR、ErbB1、人HER1)是一种细胞表面受体,是胞外蛋白配体表皮生长因子家族(EGF家族)的受体 (1)。表皮生长因子受体存在于细胞膜上,特异性配体结合引起受体激活,配体包括表皮生长因子和转化生长因子α(TGFα)。突变可以导致EGFR受体的过度表达(上调)或过度活化,这与许多癌症相关,包括肺癌、肛门癌和胶质母细胞瘤(2)。在后一种情况下经常观察到EGFR的特异性突变,又称为EGFRvIII。EGFR或家族成员的突变、扩增或失调与大约30%的上皮癌有关。EGFR突变可能导致其持续激活,造成不受控制的细胞分裂,引发癌症易感性。在一些癌症类型中鉴定到表皮生长因子受体的突变,它是扩展抗癌疗法的重要靶标(3)。
