腺激肽释放酶属于胰蛋白酶样丝氨酸蛋白酶,是大鼠垂体前叶中主要的雌激素诱导蛋白.由于雌激素也诱导腺激肽释放酶mRNA,因而基因表达增加可能介导上述诱导作用. Chlorotrianisene属于雌激素激动剂.
体外:Chlorotrianisene的ED50值降低了3.9倍.Chlorotrianisene呈浓度依赖性地抑制P388以及P388/ADR细胞生长[1].
体内:在3项实验中,Chlorotrianisene出现显著不同的激动作用.在子宫重量与垂体前叶催乳素方面,Chlorotrianisene是一种部分激动剂.相对于腺激肽释放酶,Chlorotrianisene显著地激动催乳素或子宫.此外,Chlorotrianisene对腺激肽释放酶的激动作用不同于对子宫或催乳素的激动作用[2].
临床试验:Chlorotrianisene是一种雌激素,于口服给药(48 mg/day)7天后,有效地抑制产后泌乳.Chlorotrianisene几乎没有引起症状复发以及撤退性出血,可能因为Chlorotrianisene于治疗停止后存储于人体脂肪中,随后被逐渐地释放[3].
