CCG-50014是一种有效的RGS小分子抑制剂(对RGS4的IC50值为30 nM,对RGS4的选择性是其他RGS蛋白的20多倍).RGS是GTPase活化蛋白,可结合活化异源三聚体G蛋白的Gα亚基,并能提高GTP水解速率.[1][2]
RGS(G蛋白信号调控子)可促进GTPase的alphas亚基水解能力,使G蛋白失去活性,从而抑制G蛋白信号转导.
在存活的HEK293T细胞中,CCG-50014可抑制Gαo依赖的GFP-RGS4膜易位.转染GFP-RGS4和Gαo以及对照(0.1% DMSO)的HEK293T细胞不会改变RGS4的膜定位,CCG-50014(100 μM)可逆转RGS的膜定位.[1]
