Tizoxanide是抗厌氧细菌、原生动物和一系列病毒的噻唑化物抗感染药物nitazoxanide的活性代谢物。
乙型肝炎病毒(HBV)和丙型肝炎病毒(HCV)是主要的公共卫生问题,在全世界造成超过5亿的慢性感染。 HBV和HCV都是肝脏疾病的来源,是肝细胞癌的主要风险因素。
体外:先前研究发现,tizoxanide对HBV和HCV复制都显示出有效抑制。Tizoxanide还表现出对2.2.15细胞的胞内HBV复制和胞外病毒产生的选择性抑制。Tizoxanide可以选择性降低AVA5细胞中的胞内HCV复制。此外,tizoxanide与重组人干扰素α2b(IFNα)或NS5B(HCV聚合酶)抑制剂2'-C-甲基胞苷联合用药,导致针对HCV复制的协同相互作用。此外,tizoxanide对全长基因型1a复制子的抗病毒活性与对AVA5细胞观察到的相当[1]。
体内:在免疫抑制大鼠模型中的动物研究表明,与不可吸收的sinefungin和paromomycin相比,用nitazoxanide(tizoxanide的母体药物)治疗后复发较少[2]。
临床试验:tizoxanide的母体药物Nitazoxanide目前正处于治疗慢性丙型肝炎的II期临床研究[1]。
