G-15是一种选择性的GPR30拮抗剂,Ki值为20 nM [1]。
G蛋白偶联受体30(GPR30)是一种整合膜蛋白,位于内质网中,与雌二醇和醛固酮具有高亲和性。GPR30参与多种细胞内信号通路 [1]。
G-15是一种选择性的GPR30拮抗剂,Ki值为20 nM,在浓度高达10 μM时对ERα或ERβ几乎没有结合。在仅表达GPR30的SKBr3乳腺癌细胞中,G-1或雌激素显著增加细胞内钙离子的浓度,而G15以剂量依赖的方式抑制G-1或雌激素引起的反应,IC50值分别为185和190 nM。在转染GPR30的COS7细胞中,G-15抑制雌激素和G-1对PI3K的活化以及PIP3的积累 [1]。在子宫内膜异位细胞中,G-1增加细胞增殖和Akt的磷酸化,而G-15逆转这种刺激,抑制细胞增殖,诱导Akt的去磷酸化 [2]。
在雌二醇处理的完整大鼠和去势(OVX)大鼠中,G-15损害持续转弯的延迟匹配到位置(DMP)T型迷宫任务的获得。该结果表明,GPR30在介导雌二醇对空间学习的效应中起至关重要的作用 [3]。

G-1(30 μM)在2小时内导致Akt的serine 473位磷酸化。用G-15(60 μM)预处理可防止G-1诱导的磷酸化。10 μM G-1处理72小时显著刺激细胞增殖,随后用30 μM G-15处理72小时显著逆转了这种刺激。G-15减少相对增殖。G-15以剂量依赖性方式抑制SKBr3细胞中G-1介导的钙动员,IC50大约为185 nM。在GPR30转染的COS7细胞中,G-15抑制G-1介导的PI(3)K的激活。 |
