SMER 28是一种新型的小分子增强剂,加强细胞生长抑制作用。Rapamycin靶点蛋白能调节多种细胞过程,包括自噬,因而在某些神经退行性疾病与传染病中起着保护作用。
体外:在哺乳动物细胞中,SMER 28单独诱导Rapamycin自噬,促进自噬底物清除,如A53T α-突触核蛋白和突变亨廷顿蛋白(与亨廷顿氏病和帕金森氏病相关)。在亨廷顿氏病患者的细胞中,SMER 28单独作用于Rapamycin靶点下游,减少突变亨廷顿蛋白片段的毒性[1]。
体内:以往研究表明,在具备自噬能力的小鼠胚胎成纤维细胞(MEF)(Atg5+/+)中,EGFP-HDQ74聚合物因自噬而减少。相对于未经处理的Atg5+/+细胞,未经处理的Atg5-/-细胞(自噬缺陷)具有显著增加的EGFP-HDQ74聚合物。在Atg5+/+细胞中,SMER 28显著减少EGFP-HDQ74聚合物,但Atg5-/-细胞未出现上述现象。因此,SMER 28只能在具备自噬能力的细胞中减少突变亨廷顿蛋白聚集[1]。
