MTT(3-(4,5-二甲基-2-噻唑基)-2,5-二苯基-2H-溴化四唑)是一种用于检测体外细胞增殖的试剂。
在细胞生物学中,四唑盐已被广泛用于检测从微生物细胞到哺乳动物细胞的代谢活性。哺乳动物细胞的组分研究表明,NADH(还原型吡啶核苷酸辅因子)能使大多数MTT还原,全细胞研究也进一步证明上述结论。
体外:MTT还原不仅与线粒体相关,同时也与细胞质以及非线粒体膜(包括核内体膜、溶酶体膜和质膜)相关。MTT所带的净正电荷是通过质膜电位参与其细胞摄取过程的主要因子。然而,第2代四唑染料(XTT、WST-1以及MTS)形成水溶性formazans,需要用于还原反应的中间电子受体。由于第2代四唑染料携带一个净负电荷,因而其细胞渗透性极差[1]。
体内:目前,MTT仅用于细胞体外增殖测定,暂无体内研究。
