背景

Phosphocreatine (also known as creatine phosphate) disodium salt is a phosphorylated creatine molecule that may serve as a quickly accessible reserve of high-energy phosphates in skeletal muscle, myocardium as well as the brain. Phosphocreatine can offer a phosphate group to ADP for ATP synthesis, or conversely, excess ATP can be used to convert creatine to creatine phosphate via creatine kinase. Thus, phosphocreatine disodium salt could be used as a substrate for the determination of creatine kinase, and is used to regenerate ATP during skeletal muscle contraction.

试验操作

Cell experiment:[3]

Cell lines

H9C2 cells

Reaction Conditions

5 ~ 20 mM phosphocreatine for 2 h incubation

Applications

Phosphocreatine was found to provide a significant protective effect in H9C2 cells injured by exposure to methylglyoxal. Moreover, phosphocreatine at different concentrations (5 ~ 40 mM) has no lethality on normal H9C2 cells.

Animal experiment:[3]

Animal models

Wistar male rats with type 1 diabetes mellitus

Dosage form

20 or 50 mM

Administered intravenously once daily for 12 weeks

Applications

In diabetic rats, phosphocreatine significantly reduced heart sizes, reduced blood glucose levels, and improved the striation of myocardial fibers arranged regularly in treated groups. Phosphocreatine offered a defensive impact against diabetic cardiomyopathy.
