Puromycin dihydrochloride(嘌呤霉素盐酸)是氨酰tRNA的类似物,是一种氨基核苷类抗生素,常用于筛选通过质粒转染/转 化、病毒感染等方法能表达pac基因的真核或原核多克隆或单克隆细胞。Puromycin不仅用于稳定细胞株的筛选,也用于稳定细胞株的维持。[1]。它能抑制氨酰tRNA与合成多肽的C末端结合,导致多肽链的提前终止。
嘌呤霉素(Puromycin)对各种真核细胞包括哺乳动物细胞的生长是有毒的。0.510 μg/ml嘌呤霉素可以抑制哺乳动物细胞的生长。嘌呤霉素抑制恶性疟原虫和贾第鞭毛虫生长的IC90值分别是60 μg/ml和54 μg/ml[2]。氨酰tRNA是核糖体上合成的多肽延伸的材料[2]。
不同浓度的嘌呤二盐酸可以改变嗜热四膜虫的生长速率。在第一个24小时,50 μg/ml的嘌呤霉素二盐酸盐将生长速率降低80%,但并没有完全阻断细胞生长,直至72小时,细胞数量才逐渐增加。100 μg/ml嘌呤霉素二盐酸盐完全阻断细胞生长,在第一个48小时,几乎所有的细胞死亡,48小时后存活的细胞快速增长。150 μg/ml的嘌呤二盐酸盐可完全抑制细胞生长长达72小时,在72小时时,大部分细胞死亡,存活的细胞生长。200 μg/ml嘌呤二盐酸盐48小时几乎造成所有的细胞死亡,并没有幸存细胞[2]。
在25日龄的动物中,接触嘌呤霉素盐酸盐180或120分钟可抑制后续的氨基酸转运。然而,嘌呤霉素二盐酸未能抑制50日龄动物中α氨基异丁酸的吸收[3]。

在DMSO中的溶解度大于10 mM。若配制更高浓度的溶液,一般步骤如下:请将试管置于37 °C加热10分钟和/或将其置于超声波浴中震荡一段时间。原液于-20 °C可放置数月。 |
