FURA-2AM是一种可渗透细胞的高亲和力钙离子荧光探针,是比率测量中使用最广泛的钙指示剂之一。FURA-2AM是FURA-2的酯形式,FURA-2是极性大的酸性化合物,无法进入活细胞内,在其负性基团上加上乙酰氧甲酯(AM)后,显著增加了细胞渗透性。
FURA-2AM穿透细胞膜进入活细胞后,被细胞内的酯酶去酯化变成FURA-2游离酸,后者可与胞浆内的游离Ca2+特异性地结合(结合比例为1:1)。当FURA-2结合胞质内的游离Ca2+后,表现出吸收位移,激发波长峰值从380 nm变为340 nm,而510 nm附近的发射峰值保持不变[1-4]。荧光强度与结合Ca2+的浓度存在定量关系。一般用340nm和380nm波长激发Fura-2,通过使用与两种激发对应的荧光强度比率来计算细胞内的钙离子浓度。比率测量可最大程度地减少混合物中的光漂白,荧光探针的泄漏,探针的不均匀装载和细胞厚度差异的影响,从而提供更可靠和可再现的结果。
Indo-1也是目前使用最广泛的比率测量的钙荧光指示剂。通常来讲,Fura-2 是显微镜成像的首选染料,改变激发波长比发射波长更实用。而Indo-1是流式细胞仪的首选染料,使用单个激光激发(通常是氩离子激光的351-364 nm光谱线)并检测两个发射光更为实用,当染料被Ca2+饱和时,Indo-1的最大发射值从~475 nm转变为~400 nm。Fura-2比indo-1更耐光漂白。
