伯氨喹(Primaquine)是8-氨基喹啉,在1950年作为治疗性抗疟疾药引入。从此以后,广泛使用该药剂治疗疟疾红血细胞外期。Primaquine通过与核酸结合,可阻止蛋白合成、改变脂质合成及与生物膜相互作用[1]。
体外实验:采用鸡胚细胞(CEC)模型研究primaquine对新城鸡瘟病毒复制的作用。研究发现,primaquine剂量依赖性地抑制病毒诱发的红细胞吸附,250 μg/ml的primaquine可完全抑制。研究发现,当在病毒复制周期早期加入primaquine可抑制病毒核糖核酸(RNA)的合成。然而,当在病毒复制周期后期加入该试剂可刺激RNA的合成[1]。
体内实验:将99mTc标记的primaquine脂质体静脉注射入瑞士白化小鼠体内。注射后,脂质体主要积累在肝脏中,然后是脾脏、胰腺、肺及其他。研究也表明,primaquine可阻止消灭寄生虫,通过破坏红外肝期来防止复发[2]。
临床试验:在一项双盲、随机的及安慰剂对照临床试验中,受试者每天给药氯喹与两粒磷酸伯氨喹片(30 mg) 或相应的安慰剂(2:1)。在一组100名受试者中,氯喹/伯氨喹用药具有显著的保护作用。与51名受试者的安慰剂用药组相比,氯喹/伯氨喹对88%的所有类型疟疾、89%的恶性疟疾及88%的间日疟原虫疟疾具有抑制作用[3]。
