Amphotericin B是从链霉菌(Streptomyces nodosus)菌株产生的多烯类抗真菌抗生素。IC50 =0.028-0.290μg/ml
体外实验:Amphotericin B是治疗许多危及生命的真菌感染的有效药物。在表达TLR2和CD14的细胞中,Amphotericin B诱导信号转导和炎症细胞因子释放。在表达TLR2、CD14和衔接蛋白MyD88的原代鼠巨噬细胞和人细胞系中,Amphotericin B诱导NF-κB依赖性报告子活性和细胞因子释放,而细胞缺乏任何一个均对Amphotericin B没有反应。具有TLR4突变的细胞对Amphotericin B刺激的反应比表达正常TLR4的细胞反应弱[1]。Amphotericin B可与哺乳动物膜的主要固醇,即胆固醇相互作用,由于其具有相对高的毒性,限制了Amphotericin B的有用性[2]。在悬浮于等渗蔗糖溶液中的负载KCl的脂质体中,低浓度AmB(≤ 0.1 μM)诱导极化电位,表明K+渗透。大于0.1 μM AmB允许阳离子和阴离子运动。在悬浮在等渗NaCl溶液中的LPs中, 0.05 μM AmB导致负膜电位几乎完全崩溃,表明Na+进入细胞[3]。
在体实验:在仓鼠瘙痒病模型中,Amphotericin B延长了孵育时间,降低PrPSc积累。在具有传染性亚急性海绵状脑病(TSSE)的小鼠中,Amphotericin B显著降低PrPSc含量[4]。
