天然产物安丝菌素P-3(ansamitocin P-3)是一种美登素类化合物,对人实体瘤细胞系A-549和HT-29有强效细胞毒性,ED50值分别为4 x 10-7和4 x 10-7 μg/ml [1]。
细胞毒素剂仍然是一种重要的化疗策略。在活性导向分离鉴定中,从两种藓类植物的活性甲醇提取物中取得安丝菌素P-3。此外,该化合物对几种肿瘤细胞具有极大的细胞毒性[1]。
安丝菌素P-3是一种从两种藓类植物羽藓科(即麻羽藓和狭叶牛舌藓)中以极低产率分离的美登素类化合物。对安丝菌素P-3的进一步生物学评价显示,其对人实体瘤细胞系A-549肺癌细胞、HT-29结肠癌细胞及MCF-7乳腺癌细胞有强效细胞毒性,ED50值分别为4 x 10 -7、4 x 10 -7及2 x 10 -6 μg/ml[2]。
安丝菌素P-3对部分去纤毛的四膜虫的纤毛再生有抑制作用。通过观察去纤毛不动细胞活力的恢复情况来测试四膜虫的纤毛再生。孵育60分钟后,在恢复缓冲液中95%以上的部分去纤毛的四膜虫细胞有充分能动性。1 μM的安丝菌素P-3完全抑制四膜虫的纤毛再生。此外,在2 μM的安丝菌素P-3抑制作用解除后,部分去纤毛细胞活力恢复[2]。
