BMS-794833是有效的ATP竞争性c-Met/VEGFR2双重抑制剂,其IC50值分别为1.7 nM和15 nM.BMS-794833对Ron\Axl和Flt3也具有抑制作用,其IC50值小于3 nM.
c-Met,亦称MET,是一种膜受体,对胚胎发育与伤口愈合是必不可少的.VEGFR是血管内皮生长因子的受体,属于受体酪氨酸激酶.VEGFR介导各种细胞功能,包括内皮细胞增殖\迁移\存活\管状形态发生和出芽[1].
BMS794833也抑制c-Met受体刺激的胃癌细胞系(GTL-16),其IC50值为39 nM[1].在人胃癌异种移植模型中,至少经过1个肿瘤倍增时间时,BM798433显示出大于50%的肿瘤生长抑制,同时,14天内没有出现显著毒性.在U87胶质母细胞瘤模型中,25 mg/kg BMS798433使肿瘤完全停止生长[1].
