BMS493是pan-视黄酸受体(pan-RARs)的反相激动剂。该物质可增加核共阻遏物(NCoR)与RAR的相互作用。
视黄酸受体(RAR)是一类核受体,是配体依赖的转录因子,可被9-顺式视黄酸和全反式视黄酸活化。激动配体结合RAR可引起共阻遏物解离和共激活蛋白的募集,从而促进下游靶基因转录为mRNA,最终合成蛋白质。该蛋白可控制多种生理过程,包括细胞生长、分化、存活和死亡。
在细胞培养物中,BMS493预处理MEPM细胞可阻断atRA诱导的细胞凋亡指数。从机理上说,该物质可完全抑制atRA诱导的Sub-G1分数、DNA断裂和caspase-3活化[1]。
在体内,使用BMS493抑制RAR信号可诱导程度非常高的先天性膈疝,这与取决于药物递送时间的显著的左右片面性有关[2]。
