Biotin hydrazide是一种生物素衍生物,可用作探针,用于检测蛋白质的羰基化,其是多种疾病的成分。蛋白质羰基化是一种不可逆的翻译后修饰(PTM),由活性氧(ROS)攻击、大量的脂质氧化产物(如α,β不饱和γ-hydroxyalkenals)或非酶糖化所引起,从而导致蛋白功能的损失。Biotin hydrazide是一种优选的羰基反应性探针,不需要额外的催化剂或还原剂。Biotin hydrazide在pH 5.5条件下可以完全并容易地衍生羰基,使其可用于检测生物样品中羰基化的蛋白质。

应用 |
在与生物素反应前, 100mg的人脐带透明质酸在0.1 M的 Mes溶液中(pH 5.0)至少溶解24h(4℃) 才能达到最终浓度为1 mg/ml。Biotin hydrazide 用DMSO溶解成100 mM 的储存液,然后将透明质酸溶液加入其中,使得Biotin hydrazide 溶液的终浓度为1 mM。溶于水的浓度为100 mM 的EDAC 加入到透明质酸-biotin 溶液中,使得EDAC 的终浓度为30 mM。 混合溶液4℃过夜搅拌。没有连上的生物素以及EDAC 可以用4M的盐酸胍溶液去除。透析过的生物素化的透明质酸在-20℃下能够保存几个月。 |
