BAY 41-2272是一氧化氮敏感性鸟苷酸环化酶(NO-sensitive GC)的激活剂,在存在和不存在100 nmol/L 的DEA-NO条件下,EC50值分别为0.3 μmol/L和3 μmol/L [1]。
NO敏感性GC催化cGMP的形成。它通常被认为是信号分子NO的最重要受体。NO/cGMP途径在许多生理过程中发挥作用,如血小板聚集的抑制和平滑肌的松弛[1]。
在血小板中,3 μmol/L的GSNO(次于最大有效浓度)被用来评估BAY 41-2272对NO敏感性GC的增敏作用。在缺少BAY 41-2272的情况下,此浓度下cGMP对NO的应答是微不足道的。在100 μmol/L的BAY 41-2272下,3 μmol/L的GSNO导致cGMP快速增加至高达1000 pmol/109血小板 [1]。
sGC/NO系统参与勃起功能障碍的发病机制。静脉内注射1 mg/kg 的BAY 41-2272诱导家兔产生非常弱的勃起,在10分钟时粘膜暴露的最大长度约为3 mm,且效果持续近似30分钟。SNP是NO供体。同时给予SNP可增加BAY 41-2272的效果。静脉给药0.2 mg/kg 的SNP产生了大约5-10分钟的短时间持续勃起,未覆盖阴茎粘膜的峰值长度为5 mm。IV给药1 mg/kg的BAY 41-2272,5分钟后IV给药0.2 mg/kg 的SNP,产生平均15毫米的勃起长度,比两种药物分别使用引发的勃起长度更长[2]。
