AV-412是一种EGFR和ErbB2激酶的双重抑制剂,IC50值分别为43 nM和282 nM[1].
在含有EGFR突变型的H1650和H1975细胞系中,浓度为0.1 μM的AV-412抑制EGFR的自磷酸化及随后Akt和Erk的活化.在含 ErbB2突变型的H178细胞系中,AV-412也抑制EGFR\ErbB2和ErbB3的磷酸化作用.AV-412也减少了Akt和Erk的磷酸化作用[2].
AV-412完全抑制裸鼠中H1650和H1975异种移植物的肿瘤生长.实验证明,AV-412通过抑制EGFR,抑制肿瘤生长.除此之外,AV-412对多种表达EGFR\ErbB2或两者的肿瘤模型都有抗肿瘤活性,如乳腺癌KPL-4\前列腺癌DU145和肺癌LC-376.H520肺癌由于缺少EGFR和ErbB2,所以对AV-412不敏感[2].
