AS 1949490是一种选择性的含SH2结构域的肌醇5磷酸酶2(SHIP2)抑制剂[1],作用于小鼠和人SHIP2的IC50值分别为0.34 μM和0.62 μM[2]。
SHIP2是一种胞内磷酸酶,作为一种负调节因子,在胰岛素信号中具有重要作用。由于其抑制作用,在Ⅱ型糖尿病治疗中具有潜在作用[2]。
Akt是调节胰岛素信号的关键酶。在L6肌肉细胞中,AS 1949490剂量依赖性增加胰岛素诱导的Akt磷酸化,但没有引起Akt蛋白整体水平和胎牛血清诱导的Akt磷酸化的任何相关变化。添加不同浓度AS 1949490和1 nM胰岛素培养48小时,培养基中葡萄糖的消耗呈剂量依赖性增加[2]。
在db/db糖尿病小鼠中,AS 1949490口服给药,一日两次连续7或10天,可显著减少血糖浓度(相对于vehicle组减少23%),但不影响胰岛素水平、食物摄入或体重。AS 1949490处理第10天,可显著减少血糖浓度时间曲线下的面积(AUC)和空腹血糖浓度(相对于vehicle组减少37%,时间约为30分钟)[2]。
