8-乙氧基-9-乙基腺嘌呤(ANR 94)在体外是一种腺苷受体拮抗剂。该物质的结构已公开[1]。ANR 94对人类腺苷A2A受体亚型具有高选择性和亲和性,并在单侧6-羟基多巴胺(6-OHDA)损伤的大鼠中具有高抗帕金森病活性[2]。ANR 94对腺苷A2A受体的Ki值为46 nM[1]。

在稳定转染人类重组腺苷受体的中华仓鼠卵巢(CHO)细胞中,ANR 94比ANR 82对腺苷A2A受体有更高的选择性,Ki值为46 nM [1]。ANR 94处理(0.5 mg/kg i.p. 7天)可显著防止1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)诱导的TH阳性细胞恶化(p < 0.0005)[2]。
在大鼠中,ANR 94在5 mg/kg i.p.剂量下不能改变自发运动,然而在较高的剂量(10或15 mg/kg)下,该物质可诱导运动过度。ANR 94在1 mg/kg剂量下对全身僵硬症效果较差,但在5 mg/kg下则完全有效。在重度僵住症大鼠中,在90-min的测试周期中,ANR 94在5 mg/kg i.p.剂量下可显著逆转0.2 mg/kg氟哌啶醇诱导的僵住症。从30-60 min,ANR 94效果最强。ANR 94的抗僵住症效果持续长达150 min以上。在6-OHDA损伤的大鼠中,ANR 94可显著增加l-DOPA(3 mg/kg)诱导的对侧旋转数量,这一效果可持续到120-130 min[2]。
