体外:在表达α7神经元乙酰胆碱受体(α7 nAChRs)的卵母细胞内,A-867744加强乙酰胆碱(Ach)诱发的电流,EC50值约为1 μM。在所测试的最高浓度的A-867744时,Ach诱发的电流基本上无衰减。在较低浓度的A-867744时,与另一种II型α7PAM 4-萘-1-yl-3a,4,5,9b-四氢化-3H-环戊酮-a[c]喹啉-8-磺酰胺(TQS)相比,没有证据表明有次要成分的存在[1]。
体内:对大鼠、狗及猴体内A-867744的PK性能进行评估。A-867744在动物体内的药代动力学特性以较低的血浆清除率(1.1-2.5 L/h/kg)和跨物种分布体积为特征,其末端消除相半衰期在1.0-1.7小时的范围内。大鼠内生物利用度为83%,而狗和猴的较低,分别为55%和68%。这些发现表明,A-867744呈跨物种的可接受的药代动力学特性,其脑水平足以调节α7神经元乙酰胆碱受体[2]。
临床试验:目前A-867744处在临床前发展阶段,还没有临床数据。
