8-Bromo-cAMP是一种cAMP的类似物[1]。
cAMP可以影响细胞的增殖反应,其正调节或负调节取决于细胞类型。造血细胞系中cAMP的效果是可变的。CAMP-类似物可以显著抑制促红细胞生成素刺激的增殖[1]。
通过使用特定的cDNA探针进行杂交分析发现,8-Bromo-cAMP增加编码hCGα和β亚基的mRNA的细胞含量,阻止纤连蛋白mRNA的增加。8-Bromo-cAMP也诱导AML193细胞中ERK1,2的磷酸化。8-Bromo-cAMP是AML193细胞中激活ERK1,2的试剂,这种活性不需要Shc磷酸化的参与[1,2]。
8-Bromo-cAMP增加儿茶酚胺(catecholamine)的生物合成,同时增加酪氨酸羟化酶(tyrosine hydroxylase)的活性和磷酸化。8-Bromo-cAMP通过调节mRNA的表达,促进特定蛋白合成和分泌的改变,包括纤连蛋白和hCG的亚基[2,3]。

在灭菌水中的溶解度为100 mM。若配制更高浓度的溶液,一般步骤如下:请将试管置于37 °C加热10分钟和/或将其置于超声波浴中震荡一段时间。原液于-20 °C可放置数月。 |

实验结果 |
在生长停滞的AML193细胞中,8-Bromo-cAMP显著增加DNA合成,其ED50值约为10 μM。当剂量高于2.5 μM时,8-Bromo-cAMP显示毒性作用。加入1 μM 8-Bromo-cAMP可以增强IL-3诱导的促有丝分裂反应。此外,8-Bromo-cAMP在1μM的剂量下使Erk磷酸化程度达到最高。然而,8-Bromo-cAMP对Erk磷酸化的作用短暂,其最强作用于1 ~ 5分钟内显示。 |
