组蛋白去乙酰化酶(HDACs)从组蛋白的ε-N-乙酰赖氨酸上移除乙酰基,使组蛋白更紧密地包裹DNA.DNA表达由去乙酰化和乙酰化调节.
2-hexyl-4-Pentynoic Acid属于丙戊酸(VPA)衍生物,是一种有效的HDAC抑制剂.在小脑颗粒细胞中,2-hexyl-4-Pentynoic Acid(5 μM)呈剂量依赖性地显著提升乙酰化组蛋白H3(Ac-H3)水平.与100 μM VPA(提升200% Ac-H3水平)相比,2-hexyl-4-Pentynoic Acid在50 ~ 100 μM剂量下最大程度地提升Ac-H3水平(600 ~ 700%).此外,50 ~ 100 μM 2-hexyl-4-Pentynoic Acid能完全阻止谷氨酸诱导的细胞死亡.2-hexyl-4-Pentynoic Acid还能恢复谷氨酸诱导的神经元损失.50 μM 2-hexyl-4-Pentynoic Acid抑制HDAC,从而有效地增加HSP70-1a以及HSP70-1b mRNA水平[1].
