10-DEBC hydrochloride是Akt(或称为PKB)的选择性抑制剂[1],IC50值大约为48 μM[2]。
Akt是一种丝氨酸/苏氨酸激酶。它磷酸化并灭火细胞凋亡组分,包括Caspase 9和BAD。Akt磷酸化并抑制Forkhead转录因子,并因此促进细胞的存活[3]。
在LDH和MTT测试中,单独使用10 μM的10-DEBC dihydrochloride不影响细胞活力。在顺铂处理的U251细胞中,10 μM的10-DEBC dihydrochloride消除了二甲双胍的细胞保护作用。10-DEBC dihydrochloride可以中和二甲双胍的抗凋亡活性,增加细胞膜磷脂暴露和DNA破碎至单独的顺铂作用的水平。在U251细胞中,10-DEBC dihydrochloride显著诱导活性氧的生成。在顺铂处理的细胞中,10-DEBC dihydrochloride减弱二甲双胍的抗氧化作用[1]。
在酚妥拉明处理的BAT外植体中研究10-DEBC hydrochloride的作用。10-DEBC hydrochloride对总Akt蛋白没有任何影响,抑制大约40%的Akt Ser473的磷酸化。在WT外植体中,单独的10-DEBC dihydrochloride对AMPK Ser485/491位点的磷酸化没有效果,也没有影响酚妥拉明的效果。但是,在β-AR KO外植体中,10-DEBC dihydrochloride钝化了酚妥拉明引发的AMPK的Ser485/491磷酸化的增加[4]。
